Optimization of Microwave-Assisted Extraction of Cordycepic Acid and Cordycepin from Cultured Cordyceps militaris by Response Surface Methodology

The response surface methodology was employed to optimize the integrated extraction parameters of cordycepic acid and cordycepin from cultured Cordyceps militaris (L.) link based on a single-factor experiment.

The Box-Behnken design with three independent variables i.e., microwave power (W), water/material ratio (mL/g) and extraction time (min) was used.

The experimental data obtained were fitted to a second-order polynomial equation using multiple regression analysis.

The three-dimensional response surface plot and contour plot derived from the mathematical models were applied to determine the optimal conditions.

The optimum extraction condition was obtained as follows: microwave-assisted extraction, microwave power of 649.33 W, extraction time of 5.74 min, water/material ratio of 38. 99 mL/g and extraction number of three.

The yield of cordycepic acid and cordycepin were 2.47 and 0.79 %, respectively.

Under these conditions, the experimental values of 3.12 and 0.75 % well agreed with those predicted by the model.
